    Abstract [sv]

    I slutet av förra året publicerade livsmedelsverket en rapport om Listeria monocytogenes i kyld och ätfärdig mat. Författarna har studerat denna livsmedelsinfektion under lång tid och jämför här både äldre och nyare studier med den aktuella rapporten. Resultaten pekar på att listeriabakterier fortfarande är vanligt förekommande i kallrökt och gravad lax.

    SENSMORAL

    Vakuumförpackade kallrökta eller gravade laxprodukter bör åsättas en kortare Bäst-före-tid. Om konsumenterna konsekvent undviker att köpa produkter som närmar sig Bäst-före-dagen kanske vi automatiskt får en självsanering.

    Abstract [en]

    To stem the tide of food-borne illnesses, it is important to understand the source of the problem. This volume examines pathogenic bacteria, viruses, protozoan parasites, moulds, and mycotoxins in food. It discusses food-associated antimicrobial resistance and lessons learned from an actual food-borne outbreak. The book also explores clinical aspects of food-borne illness. The book is intended for medical science students. It will also be useful as a reference book for scientists, technologists, and inspectors in public health-related fields who regularly deal with issues related to food microbiology, food-borne infections and intoxications, and food safety.

    The first lesson learned from this outbreak was that vacuum-packed rainbow trout is not only an excellent medium for the growth of Listeria monocytogenes, but may also cause human listeriosis. Another lesson is that one single fish processing plant may spread multiple clonal types of L. monocytogenes by selling contaminated products to consumers. Thus, when investigating fish-borne outbreaks of listeriosis one should identify and type several isolates of L. monocytogenes from each food and environmental sample, since multiple clonal types might be present. The outbreak described in this paper involved at least eight human cases, three clonal types of L. monocytogenes, and lasted for 11 months. During the outbreak investigation, L. monocytogenes was also isolated from another brand of rainbow trout found in the refrigerator of one of the patients. These latter isolates belonged to a clonal type not associated with the outbreak. However, this clonal type is of considerable interest since it has been associated with foodborne outbreaks of listeriosis in several countries, and is also the second most common clonal type among human clinical isolates of L. monocytogenes in Sweden. Besides the described outbreak, it is likely that vacuum-packed, cold-smoked and gravad rainbow trout have been involved in additional cases of foodborne listeriosis in Sweden.

    Six culture media were compared with blood agar regarding colony counts and colony diameters of 53 Staphylococcus aureus strains. There were no statistically significant differences between the media. The forced differentiation of the results via cluster analysis, however, gave some indications that differences existed. In terms of colony counts, LSM, BP Oxoid and BP Oxoid + pp gave results similar to those given by blood agar. Colony diameter seemed to be a doubtful measure of the media's suitability and none of the six media showed similar diameter values to those of blood agar. As regards the appearance of the S. aureus strains on the six media, BP BBL and LSM corresponded the most closely to the inventors descriptions. The selectivities of the media were tested by cultivating 102 food samples from various sources. The most favourable retardation against micro-organisms other than S. aureus was shown by BP BBL, KRANEP and LSM. In terms of all tests performed, BP BBL was the most satisfactory medium for isolating S. aureus. PCVJ was the poorest of all media used in this study.

    Abstract [en]

    Cheeses were made of heat-treated goat milk inoculated with large numbers of a histamine-producing strain of Streptococcus faecium or a non-histamine-producing strain of S. faecalis. Every second week during ripening (91 days) the cheeses were sampled for histamine analyses and bacteriological analyses. The numbers of enterococci remained high throughout the whole period of investigation and the maximum amount of histamine detected was 8.2-mu-g/g in one of the cheeses. The enterococci seem to have no relevance from a histamine intoxication point of view in cheeses of this kind.

    Abstract [en]

    Two variants of Listeria monocytogenes were isolated from blood cultures from each of two patients with listeriosis. Each variant displayed a two-band difference in DNA profile from the other by pulsed-field gel electrophoresis. Although this difference in profile is insufficient to distinguish clearly between the variants, the possibility of co-infection with different strains of L. monocytogenes needs to be considered. We suggest that more than one colony should be selected for molecular typing to aid interpretation during investigation of the sources and routes of Listeria infection.

    Abstract [en]

    Isolates of Listeria monocytogenes saved from outbreaks of listeriosis, cases of sporadic listeriosis, and similar events do not always belong to a solitary genetic variant. Variants of the same strain may have evolved from a unique clone, and plasmid loss or gain and phage-mediated genetic changes are suggested as the main mechanism. Some of these reports are summarized in this short communication.

    By using pyrosequencing (i.e., sequencing by synthesis) 106 strains of different serovars of Listeria monocytogenes were rapidly grouped into four categories based on nucleotide variations at positions 1575 and 1578 of the inlB gene. Strains of serovars 1/2a and 1/2c constituted one group, and strains of serovars 1/2b and 3b constituted another group, whereas serovar 4b strains were separated into two groups.

    Altogether, 100 strains of Listeria monocytogenes serovar 1/2a isolated from humans, animals, food, and the environment were typed by a combination of PCR and restriction enzyme analysis (REA). A PCR product of 2,916 bp, containing the downstream end of the gene inlA (955 bp), the space between inlA and inlB (85 bp), and 1,876 bp of the gene inlB, was cleaved with the enzyme AluI, and the fragments generated were separated by gel electrophoresis. By this method two different cleavage patterns were obtained. Seventy of the 100 strains shared one restriction profile, and the remaining 30 strains shared the second one. No relation was found between the types differentiated by PCR-REA and the origins of the strains.

    Faecal samples from 102 clinically healthy dairy cows, representing 34 farms in the Swedish province of Uppsala, were analysed for the presence of Listeria spp. using an enrichment procedure. Listeria monocytogenes was isolated from six (6%) and L. innocua from 2 (2%) cows. From each of the 6 samples positive for L. monocytogenes, 5 isolates were further characterised by restriction enzyme analysis using the 3 enzymes Apa I, Sma I, and Asc I, followed by pulsed-field gel electrophoresis. Three of the L. monocytogenes positive cows lived at the same farm, and they all harboured the same clonal type. One of these 3 cows also harboured a further clonal type of L. monocytogenes. The fact that one of the cows harboured 2 different clonal types of L. monocytogenes is important from an epidemiological point of view when routes of infection are to be investigated.

    The International Dairy Federation Revised Provisional IDF Standard 143:1990 (1990) is compared with the ISO Draft International Standard ISO/DIS 11290-1 (1995) for detection of Listeria monocytogenes in inoculated samples of the blue veined cheeses Gorgonzola and AG delost (Swedish blue mould cheese) and in non-inoculated market cheeses of mostly soft mould cheese. The ISO method detected L. monocytogenes signi"cantly more frequently than the IDF method when inoculated Gorgonzola samples were analysed. For inoculated AG delost, and non-inoculated market cheeses the methods were equally good. The selective isolation media PALCAM and Oxford Listeria selective agar, were compared for recovery of Listeria spp. Less growth of non-Listeria colonies was obtained on PALCAM agar compared to Oxford Listeria selective agar especially after the primary enrichment step of the ISO method. Consequently, it was easier to identify and isolate growth of Listeria species from the cheeses on the PALCAM agar.

    The incidence of Listeria species in raw whole milk from farm bulk tanks and from raw milk in storage at a Swedish dairy plant was studied. Listeria monocytogenes was found in 1.0% and Listeria innocua was found in 2.3% of the 294 farm bulk tank (farm tank) milk specimens. One farm tank specimen contained 60 CFU of L. monocytogenes ml-1. L. monocytogenes was detected in 19.6% and L. innocua was detected in 8.5% of the milk specimens from the silo receiving tanks at the dairy (dairy silos). More dairy silo specimens were positive for both Listeria species during winter than during summer. Restriction enzyme analysis and pulsed-field gel electrophoresis were applied to 65 isolates of L. monocytogenes, resulting in 16 different clonal types. Two clonal types were shared by the farm tank milk and the dairy silo milk. All except one clonal type belonged to serovar 1/2a. In the dairy silo milk five clonal types were found more frequently and for a longer period than the others. No Listeria species were found in any other samples from the plant.

    Microbiological and sensory methods were used to analyze 22 soft cheeses, of which 19 were made from raw milk, one was made from both raw and pasteurized milk and two were made from pasteurized milk. Moderate correlations (r-value 0.5–0.6 and p-value <0.01–0.05) were found between the levels of Enterobacteriaceae 37°C and the intensity of the sensory characteristics “bitter”, “metallic”, “pungent”, “manure” and “ammonia”. The present study indicates that it is possible to predict high levels of Enterobacteriaceae in soft cheeses made from raw milk using only the human senses (odor and taste).
